Identification of bacillary agent of food poisoning and corruption foodstuffs PP051
Purpose and areas of usage
The development is intended to accelerated identification of agents causing food poisoning and food corruption. The proposed methodology can be used in the food industry, biology, molecular biology, medicine, microbiology in laboratories equipped with appropriate equipment and skilled personnel.
Gist of development
The proposed development is based on polymerase chain reaction (PCR). Molecular genetic diagnostics with selected conditions and pairs of primers allows to accelerate the process. In comparison with known methods of phenotypic diagnosis of regulated bacilli 2 - 3 times due to detection of specific genes with simultaneous determination of entero - and emetic toxicity. Due to the identification of regulated microorganisms, in particular B. cereus, as well as bacilli causing food spoilage (R. rolymyxa, R. macerans), it becomes possible to carry out a comprehensive assessment of several bacilli contaminants. According to the developed schemes that can be implemented for different product groups
Scheme of the accelerated method for the identification of toxigenic Bacillus cereus, Paenibacillus polymyx and Paenibacillus macerans in plant material and its processing products:
Sampling => Preparation of food samples for identification of microorganisms => Selection of DNA of bacterial cells => Realization multiplex PCR for molecular genetic diagnostics Bacillus cereus, Paenibacillus polymyxa and Paenibacillus macerans => electrophoresis distribution of amplification products => Visualization of amplification products
Main advantages of development
The proposed approach and methodology allow to accurately and quickly identify potentially dangerous objects with microbiological contaminants. It is important for the determination of the safety of food products and food raw materials and ecological safety, as well as monitoring of the quality of food systems
The development can be used on modern food factories with laboratories in regional veterinary inspections for the control of safety and quality in order to preserve and improve the health of the population of the country.
